Abstract:A series of experiments (including culture medium, the culture time of mycelium, kinds of osmotic stabilizer and its concentration, the proper enzymes and their proportion, enzyme hydrolyzing time) were conducted to study the optimun condition of Monascus rubber protoplast. Results indicated that suitable culture medium was PDA, the culture time of mycelium was 60-day-age, the proper enzymes were snail enzyme and cellulose enzyme, the proper concentration was 1.5% (snail enzyme: cellulose enzyme=7:3), and the osmotic stabilizer was the 0.6mol/L MgSO4. Under the above conditions, the protoplasts formation was 1.61×107/ml, and the regeneration rate was 8.49%.
