Abstract: This article in order to improve the strain Bacillus subtilis var. niger ATCC 9372, spores formation rate bacillus subtilis agents in the application of life. Therefore, study the Bacillus subtilis growth curve, the source of carbon, nitrogen source, inorganic salt and growth factors of its total MnSO4·H2O count and spores formation rate. Through the growth curve is 530nm for under test in the wavelength. Bacillus subtilis is through the method of total into training, plate counting income. Bacillus subtilis is the spores in bacteria in water sample after 20min at 80℃, through cultivating, plate counting into law. First to test the single factor experiment, choose through spores are formed rate is high, carbon source (glucose and galactose) and nitrogen source (Peptone and beef extract) and NaCl and the optimal concentration of Mn2+, finally through orthogonal experiment. Final results show Bacillus subtilis ATCC937214h in its maximum growth, while the most suitable for medium composition Peptone 5g/L, 5g/L, lactose NaCl5g/L, and the concentration of Mn2+ is 2.4 mmol /L.
